目的:建立高效液相色谱(HPLC)法测定不同品种怀地黄中梓醇和毛蕊花糖苷的含量,采用星点设计-效应面法优化提取工艺。方法:建立HPLC法,甲醇回流测定梓醇和毛蕊花糖苷,Agilent ZORBAX SB-Aq色谱柱(4.6mm×250mm,5μm),流速1ml/min,柱温30℃,以乙腈~0.1%磷酸溶液(1:99)为流动相,检测波长210nm,测定梓醇含量,以乙腈~0.1%醋酸溶液(16:84)为流动相,检测波长334nm,测定毛蕊花糖苷含量。结果:不同品种怀地黄中梓醇和毛瑞花糖苷含量存在明显差异,梓醇含量最高品种为85-5,达6.58%,毛蕊花糖苷含量最高品种为9302,达0.2479%。结论:不同品种怀地黄中梓醇和毛蕊花糖苷含量不同,星点设计-效应面法优化提取工艺,方法简便且预测性好。
OBJECTIVE: To establish a method for the determination of catalpol and verbascoside in different varieties of Rehmannia glutinosa Franch.by high performance liquid chromatography (HPLC), and optimize the extraction process by using the star-point design-response surface method. Methods: HPLC and methanol reflux were used to determine catalpol and verbascoside on an Agilent ZORBAX SB-Aq column (4.6 mm × 250 mm, 5 μm) at a flow rate of 1 ml / min. The column temperature was set at 30 ℃ with acetonitrile ~ 0.1% phosphoric acid solution 99) as the mobile phase. The detection wavelength was 210 nm. The catalpol content was determined. Acetonitrile to 0.1% acetic acid solution (16:84) was used as the mobile phase. The detection wavelength was 334 nm. The content of verbascoside was determined. Results: The contents of catalpol and sorghum in different varieties of Rehmannia glutinosa were significantly different. The highest content of catalpol was 85-5, up to 6.58%, and the highest species of verbascoside was 9302, reaching 0.2479%. Conclusion: The contents of catalpol and verbascoside in different varieties of Rehmannia glutinosa are different, and the star design-response surface method is the best method for the extraction of catgut, and the method is simple and predictable.
