目的:从氧化应激及线粒体损伤方面探讨莪术油注射液对人卵巢癌SKOV3细胞胀亡可能的作用机制。方法:以莪术油注射液作用人卵巢癌SKOV3细胞后,通过全自动生化分析仪测定细胞上清液乳酸脱氢酶(LDH),荧光染料2’,7’-二氯荧光素乙二酯(DCFH-DA)、10-壬基吖啶橙(NAO)结合荧光化学发光仪分别检测细胞内活性氧(ROS)及线粒体心磷脂(MTCL)水平。结果:莪术油注射液作用SKOV3细胞后可使细胞上清液LDH水平上升,细胞内ROS生成增加,MTCL氧化损伤。结论:氧化应激诱导SKOV3细胞线粒体损伤,可能是莪术油注射液致SKOV3细胞胀亡的作用机制之一。
OBJECTIVE: To explore the possible mechanism of the effect of Zedoary Turmeric Oil Injection on the expansion and destruction of human ovarian cancer SKOV3 cells from oxidative stress and mitochondrial damage. Methods: Human ovarian cancer cell line SKOV3 was treated with Curcuma oil injection and the contents of lactate dehydrogenase (LDH), fluorescent dye 2 ’, 7’-dichlorofluorescein ethylene glycol ester ( DCFH-DA) and 10-nonyl acridine orange (NAO) combined with fluorescence chemiluminescence instrument were used to detect intracellular reactive oxygen species (ROS) and mitochondrial cardiolipin (MTCL) levels respectively. Results: After treated with zedoary turmeric oil, SKOV3 cells increased the level of LDH in the cell supernatant, increased ROS production in the cells and oxidative damage in MTCL. Conclusion: Oxidative stress induced mitochondrial injury in SKOV3 cells may be one of the mechanisms of the expansion and contraction of SKOV3 cells induced by zedoary turmeric oil.
