Objective To investigate the changes of IL-23p19 mRNA and IL-23 in peripheral blood mononuclear cells before and after treatment of type 2 diabetes mellitus (T2DM). Methods Fasting blood glucose (FBG) levels were measured in selected subjects. Fifty-eight patients with type 2 diabetes mellitus (28 newly diagnosed diabetic patients and 30 treated patients) and 26 healthy controls were detected by reverse transcription-polymerase chain reaction (RT-PCR) (Control group), the level of IL-23 in the plasma was detected by enzyme-linked immunosorbent assay (ELISA). Results The expression of IL-23 mRNA in the newly diagnosed diabetic group was higher than that in the untreated group and the control group (P <0.05). The plasma level of IL-23 in the newly diagnosed diabetic group was significantly lower than that in the untreated diabetic group and the control group Significance (P <0.05). Conclusion The levels of IL-23 and p19 mRNA in type 2 diabetes mellitus were significantly higher than those in the untreated group and control group before treatment. IL-23 may be related to the pathogenesis and progression of type 2 diabetes mellitus.
