目的采用UPLC-Q-TOF/MS(超高效液相色谱联合飞行时间串联质谱)对埃博霉素B纯化产品及黏细菌发酵液提取物进行分析。方法运用ACQUITY HSS T3超高效液相色谱柱(2.1 mm×50 mm,1.8μm),以甲醇-0.1%甲酸溶液为流动相梯度洗脱;质谱使用ESI离子源,负离子模式下采集数据。结果运用UPLC-Q-TOF/MS方法可以检测到埃博霉素B纯化产品中的微量杂质以及发酵液提取物中主要产物。结论 UPLC-Q-TOF/MS方法操作简单、快捷,可作为埃博霉素B的检测方法。
OBJECTIVE To analyze the purified product of epothilone B and the myxobacteria fermentation broth extract by UPLC-Q-TOF / MS (ultra performance liquid chromatography coupled with time of flight mass spectrometry). Methods The mobile phase was eluted with ACQUITY HSS T3 column (2.1 mm × 50 mm, 1.8 μm) using methanol-0.1% formic acid as mobile phase. The mass spectra were acquired with ESI ion source and negative ion mode. Results The trace impurities in the epothilone B purified product and the major product in the fermentation broth extract can be detected using the UPLC-Q-TOF / MS method. Conclusion The UPLC-Q-TOF / MS method is simple and rapid and can be used as a detection method for epothilone B.
