76只健康豚鼠用于观察乙基西梭霉素(Netilmicin,NM)对耳蜗的毒性作用。动物分2组,第1组54只豚鼠作急性试验,测定1次肌注乙基西梭霉素后在血液和内耳外淋巴中的药物动力学参数。另1组22只豚鼠等分成2亚组,分别连续肌肉注射NM15Omg/kg和庆大霉素(GM)125mg/kg14d,观察它们的耳蜗动作电位(CAP)阈值和耳蜗病理的改变。结果显示:两种药物在外淋巴中的药物动力学改变十分相似,而NM的耳毒性明显低于GM。提示NM的轻微耳毒性作用在于其本身药物的特点而非进入外淋巴的量较GM少,展现了NM良好的应用前景。
76 healthy guinea pigs were used to observe the toxic effects of Netilmicin (NM) on the cochlea. Animals were divided into two groups, the first group of 54 guinea pigs for acute test, determination of intramuscular injection of fosfomycin in blood and inner ear lymphatic pharmacokinetic parameters. Another group of 22 guinea pigs were equally divided into 2 subgroups. NM15Omg / kg and 125mg / kg gentamycin (GM) 125mg / kg were injected intramuscularly continuously to observe their changes of cochlear action potential (CAP) and cochlear pathology. The results showed that the pharmacokinetic changes of the two drugs in the perilymph were very similar, whereas the ototoxicity of NM was significantly lower than that of GM. It is suggested that the mild ototoxicity of NM lies in the fact that NM is less than GM in its own characteristics rather than entering the perilymph, demonstrating the good application prospect of NM.
