目的研究美多芭与安坦摄入后帕金森病(parkinson’s disease,PD)大鼠行为学及大脑5-羟色胺(5-hydroxytryptamine,5-HT)表达的改变,探讨药物治疗对PD抑郁的影响。方法雄性成年Wistar大鼠30只,随机分为空白对照组与假手术组;PD大鼠45只,随机分为PD组、安坦组和美多芭组,进行行为学、体质量、糖水偏爱测试判断抑郁情况,免疫组织化学检测海马与额叶5-HT的表达。结果 PD组、安坦组和美多芭组平均体质量和糖水偏爱度明显低于空白对照组与假手术组,美多芭组与安坦组糖水偏爱度明显低于PD组(P<0.05),美多芭组大鼠额叶与海马5-HT免疫反应阳性神经细胞数较其他组显著减少(P<0.05)。结论 PD大鼠可表现抑郁,美多芭与安坦治疗加重PD的抑郁症状。美多芭导致额叶与海马5-HT降低,可能与抑郁发生有关。
Objective To investigate the changes of behavior and the expression of 5-hydroxytryptamine (5-HT) in Parkinson’s disease (PD) rats after metoprazole and anantin exposure and to explore the effect of drug treatment on PD depression . Methods Thirty male adult Wistar rats were randomly divided into blank control group and sham operation group. Forty-five PD rats were randomly divided into PD group, Antu group and metoprolol group. The behavioral, body weight, glycemic predisposition test Determine the depression, immunohistochemical detection of hippocampus and frontal 5-HT expression. Results The average body weight and glycemic preference of PD group, Avastan group and metoprolol group were significantly lower than those of blank control group and sham operation group (P <0.05) Compared with other groups, the number of 5-HT immunoreactive nerve cells in the frontal lobe and hippocampus in the Madopar group was significantly decreased (P <0.05). Conclusion PD rats can show depression, and the treatment of dendrites and amphetamines aggravates the depressive symptoms of PD. Medopa causes a decrease in 5-HT in frontal lobe and hippocampus, which may be related to depression.
