目的:探讨心脏营养素-1(CF-1)及其受体亚基gp130,白血病抑制因子受体-β(LIFR-β)在失神经骨骼肌中的表达规律。方法:选用11组Swiss小鼠,每组10只,切断其右侧胫神经,分别于不同时间点取失神经支配的腓肠肌,用Northern blot法测定肌肉中CT-1和LIFR-β,gp130的mRNA含量,探讨三者的表达规律。结果:正常肌肉组织中CT-1的表达量较高,而2个受体亚基只有低水平表达;随着骨骼肌失神经支配时间的延长,肌肉细胞中CT-1的表达进行性下降:LIFR-β和gp130均为先增高后降低,但前者的表达高峰出现在失神经支配后24h,而后者则在神经切断后1wk表达最高。结论:骨骼肌细胞内源性CT-1表达不足,对肌肉的保护作用减弱可能是失神经肌肉早期萎缩的一个重要因素。受体gp130/LIFR-β的表达增高为进一步研究应用外源性CF-1来防治失神经肌肉萎缩提供了理论依据。
Objective: To investigate the expression of cardiotrophin-1 (CF-1) and its receptor subunits gp130 and leukemia inhibitory factor receptor-β (LIFR-β) in denervated skeletal muscle. Methods: The 11 groups of Swiss mice were selected, 10 in each group. The right tibial nerve was cut off, the denervated gastrocnemius was taken at different time points. The expression of CT-1, LIFR-β, gp130 mRNA content, to explore the expression of the three rules. Results: The expression of CT-1 in normal muscle tissue was high, while the expression of two receptor subunits was only low. With the prolongation of skeletal muscle denervation, the expression of CT-1 in muscle cells decreased progressively: Both LIFR-β and gp130 firstly increased and then decreased, but the former peaked at 24 h after denervation and the latter peaked at 1 wk after nerve denervation. Conclusion: Insufficient expression of endogenous CT-1 in skeletal muscle cells and the weakening of muscle protective effect may be an important factor in the early atrophy of muscle denervation. The higher expression of receptor gp130 / LIFR-β may provide a theoretical basis for further study of exogenous CF-1 for prevention and treatment of denervated muscle atrophy.
