目的:探讨E盒结合锌指蛋白1(ZEB1)在尿道下裂患者包皮组织中的表达,分析ZEB1 mRNA和蛋白的表达水平与尿道下裂的关系。方法:实验组和对照组的包皮组织来自于37例尿道下裂修复术患儿和11例包皮环切术包皮过长患儿,37例尿道下裂根据尿道口的位置归类为24例轻中度尿道下裂和13例重度尿道下裂,通过免疫组化和RT-PCR法分别对ZEB1的蛋白和mRNA表达水平进行检测。结果:ZEB1蛋白阳性表达率在轻中度尿道下裂组为75.0%(18/24)、重度尿道下裂组为100%(13/13),明显高于对照组的9.1%(1/11),且两两比较均具有统计学意义(P均<0.05);RT-PCR显示ZEB1 mRNA表达在对照组,轻中度、重度尿道下裂组织中的相对累计光密度值比分别为(0.67±0.21)、(0.81±0.24)、(1.55±0.29),除对照组和轻中度尿道下裂组差异无统计学意义外(P=0.64),对照组和轻中度尿道下裂组分别和重度尿道下裂组比较差异均有统计学意义(P均<0.05)。结论:ZEB1在尿道下裂患儿的包皮组织中的表达明显上调,且与尿道下裂的严重程度相关,提示ZEB1基因表达上调可能导致尿道下裂的形成。
OBJECTIVE: To investigate the expression of E box combined with ZEB1 in the foreskin of hypospadias patients, and analyze the relationship between ZEB1 mRNA and protein expression and hypospadias. Methods: The foreskin in the experimental group and the control group were from 37 cases of hypospadias repair and 11 cases of circumcised circumcised children, and 37 cases of hypospadias were classified as 24 cases according to the location of the urethra Moderate hypospadias and 13 cases of severe hypospadias. The protein and mRNA expression levels of ZEB1 were detected by immunohistochemistry and RT-PCR respectively. Results: The positive rate of ZEB1 protein was 75.0% (18/24) in mild-moderate hypospadias and 100% (13/13) in severe hypospadias, which was significantly higher than that in control group (9.1%, 1/11 (P <0.05). The relative optical density values of ZEB1 mRNA expression in the control group, mild to moderate and severe hypospadias tissues were (0.67 ± 0.21), (0.81 ± 0.24) and (1.55 ± 0.29), respectively, except for the control group and the mild to moderate hypospadias group (P = 0.64), the control group and the mild to moderate hypospadias group And severe hypospadias group were statistically significant differences (P all <0.05). Conclusion: The expression of ZEB1 in the foreskin of children with hypospadias was significantly up-regulated and correlated with the severity of hypospadias. It suggested that the up-regulation of ZEB1 may result in the formation of hypospadias.
