目的 :探讨调心方对氧化损伤型类AD大鼠空间学习记忆能力、脑线粒体呼吸功能及其酶活性的影响。方法 :采用二羟延胡索酸 (DHF)加FeCl3 ADP左侧脑室注射 ,造成大鼠氧化损伤型类AD大鼠模型 ;Morris水迷宫法观察大鼠空间学习记忆能力 ;Clark氧电极法测定脑线粒体呼吸功能各项指标 ,包括R3、R4、RCR、P O、OPR及琥珀酸脱氢酶、NADH脱氢酶、CytC氧化酶活性变化及调心方的作用。结果 :与对照组比较 ,模型鼠空间学习记忆能力、脑线粒体呼吸功能和CytC氧化酶活性显著下降 ,调心方对模型鼠上述指标有明显改善作用。结论 :调心方具有改善氧化损伤型类AD大鼠空间学习记忆障碍提高脑线粒体呼吸功能和呼吸链复合物酶活性作用 ,可能与其对抗自由基氧化损伤作用有关
Objective: To explore the effect of Tiaoxin recipe on spatial learning and memory abilities, brain mitochondrial respiratory function and enzyme activities in oxidative injury-like AD rats. METHODS: Rat model of oxidative injury was induced by injection of dihydroxyfumarate (DHF) plus FeCl3 ADP in the left ventricle; Morris water maze was used to observe spatial learning and memory ability; Clark oxygen electrode method was used to determine mitochondrial respiratory function. Various indicators, including R3, R4, RCR, PO, OPR, and succinate dehydrogenase, NADH dehydrogenase, CytC oxidase activity changes and the role of heart-regulating side. RESULTS: Compared with the control group, the spatial learning and memory abilities, brain mitochondrial respiratory function and CytC oxidase activity of the model rats were significantly decreased. The Tiaoxin Recipe had a significant effect on improving the above-mentioned indexes of the model rats. Conclusion : Tiaoxin Recipe can improve the spatial learning and memory impairment of oxidative injury-like AD rats and increase the brain mitochondrial respiratory function and respiratory chain complex enzyme activity, which may be related to its anti-free radical oxidative damage
