目的采用HPLC-CAD法研究巴戟天Morindae Officinalis Radix炮制前后寡糖类成分的HPLC-CAD指纹图谱变化,为科学评价和控制其质量提供可靠依据。方法采用Shodex Asahipak NH2(250 mm×4.6 mm,5μm)色谱柱,乙腈和水为流动相,梯度洗脱,柱温为25℃,进样量为10μL。CAD参数:气压241.36 kPa,量程100 pA。结果巴戟天不同炮制品的HPLC指纹图谱共有峰特征明显,不同炮制品指纹图谱有差异。炮制后峰面积显著升高,表明巴戟天经炮制后,寡糖的量明显增加,炮制品共确定了10个共有峰。结论建立了巴戟天寡糖类成分的HPLC特征图谱检测方法,可用于巴戟天及其炮制品的质量控制。
OBJECTIVE To study HPLC-CAD fingerprints of oligosaccharides in Morinda Officinalis Radix before and after processing by HPLC-CAD method, and to provide a reliable basis for the scientific evaluation and quality control. Methods A Shodex Asahipak NH2 (250 mm × 4.6 mm, 5 μm) column was used. The mobile phase consisted of acetonitrile and water. The gradient elution was carried out. The column temperature was 25 ℃ and the injection volume was 10 μL. CAD parameters: air pressure 241.36 kPa, range 100 pA. Results Morinda officinalis different products HPLC fingerprint common peak characteristics, different fingerprints have different products. After processing the peak area increased significantly, indicating that Morinda officinalis after processing, the amount of oligosaccharides increased significantly, processed products identified a total of 10 common peaks. Conclusion The HPLC method was established for the determination of the constituents of Morinda officinalis oligosaccharides. It can be used for the quality control of Morinda officinalis and its processed products.
