目的 :探讨多个 STR基因座在异基因骨髓移植植活诊断中的应用。方法 :分别复合扩增各病例组常染色体 1 3个STR基因座和 Amelogenin基因座 ,以及Y染色体 6个 STR基因座。扩增产物进行 Gene Scan扫描 ,Genotyper分型。结果 :病例 1、2、3移植后呈完全嵌合状态 ,提示移植成活。病例 4在第一次移植未成活后 ,行二次移植后呈完全嵌合状态。结论 :常染色体 1 3个 STR基因座可以很好地应用于异基因骨髓移植植活诊断的检测。Y- STR基因座做为植活诊断的证据具有一定的局限性。
Objective: To investigate the application of multiple STR loci in the diagnosis of allogeneic bone marrow transplantation. Methods: A total of 13 STR and Amelogenin autosomal loci and 6 STR loci of Y chromosome were amplified by multiplex amplification. Amplification products were scanned by Gene Scan and genotyped. Results: Cases 1, 2 and 3 showed complete chimerism after transplantation, suggesting that the transplantation survived. Case 4 after the first transplant did not survive, after the second transplant was completely chimerism. CONCLUSION: The 13 STR loci of autosomal chromosomes can be applied to the diagnosis of allogeneic bone marrow transplantation. Y-STR locus as evidence of implantation diagnosis has some limitations.
