目的构建多层次指标评价模型以优选红花通络方(HTP)最佳提取工艺路线。方法选取有效成分、杂质控制、行为学、组织病理学、免疫监测作为准则层,方中代表性成分羟基红花黄色素A、淫羊藿苷的提取率,出膏率,大鼠机械性刺激痛觉超敏反应、机械性刺激痛觉过敏反应的缩足百分率,背根神经节细胞偏心核比率,足底P物质量作为指标层,运用模糊层次分析法(FAHP)为各指标赋权,综合评价HTP各提取工艺路线。结果准则层指标权重(ω)由高到低为免疫监测(ω=0.245)、组织病理学(ω=0.23)、有效成分和行为学(ω=0.20)、杂质控制(ω=0.125)。最佳工艺路线为红花60℃温浸、淫羊藿等水回流提取、桂枝80%乙醇回流提取。结论以FAHP为赋权方法,有效成分指标结合药效学指标和杂质控制作为评价指标来优选提取工艺路线的方法,既能保留复方有效合理的本质,又能满足现代制剂研发的要求。
Objective To establish a multi-level index evaluation model to optimize the extraction process of Honghua Tongluo Formula (HTP). Methods The effective components, impurity control, behavior, histopathology and immune monitoring were selected as the guidelines. The extraction rates of hydroxyl safflower yellow A, icariin, the rate of ointment, the mechanical stimulation of rats Hyperalgesia hyperalgesia, mechanical reduction of hyperalgesia in hyperalgesic hyperalgesia, eccentric nucleus ratio of dorsal root ganglion cells, and content of plantar P substance as index layer, using fuzzy analytic hierarchy process (FAHP) HTP each extraction process route. Results The weights (ω) in the guideline layer ranged from immune monitoring (ω = 0.245), histopathology (ω = 0.23), active ingredient and behavior (ω = 0.20) and impurity control (ω = 0.125). The best technological route is safflower 60 ℃ temperature, Epimedium water reflux extraction, Guizhi 80% ethanol reflux extraction. Conclusion FAHP as the empowerment method, the effective composition of the index combined with pharmacodynamic indicators and impurity control as an evaluation index to optimize the extraction process route, both to retain the compound effective and reasonable nature, but also to meet the requirements of modern formulations research and development.
