目的:观察急性多发脑梗死大鼠海马MARCKS mRNA表达的动态变化,探讨海马MARCKS基因表达动态变化与缺血损害的关系。方法:采用改良Kaneko法复制急性多发脑梗死模型,观察造模后大鼠的神经功能缺损评分改变,应用组织病理学方法观察模型1 h、6 h2、4 h7、2 h、7 d组缺血损害改变,并应用半定量PCR方法检测缺血各时点海马MARCKS mRNA表达水平。结果:造模后大鼠有不同程度的神经功能缺损表现,海马组织病理学改变以24 h后为著。急性缺血海马MARCKS mRNA均为过高表达,1 h开始升高,7 d最高,呈动态变化。结论:急性多发脑梗死大鼠海马MARCKS mRNA的表达异常升高,而MARCKS mRNA的过表达与海马缺血损害的发生密切相关。
Objective: To observe the dynamic changes of MARCKS mRNA expression in the hippocampus of acute multiple cerebral infarction rats, and to explore the relationship between the dynamic changes of MARCKS gene expression and ischemic damage in the hippocampus. Methods: The model of acute multiple cerebral infarction was duplicated by modified Kaneko method. The changes of neurological deficit score in rats were observed. Histopathological examination was used to observe the ischemia of models at 1 h, 6 h, 2 h, 4 h, 7 h, 7 d The changes of MARCKS mRNA expression in the hippocampus were detected by semi-quantitative PCR. Results: After modeling, rats had different degrees of neurological deficits. The histopathological changes of hippocampus were observed after 24 h. Acute ischemic hippocampal MARCKS mRNA were overexpressed, 1 h began to rise, 7 d highest, showing dynamic changes. Conclusion: The expression of MARCKS mRNA in the hippocampus of rats with acute multiple cerebral infarction is abnormally elevated. The overexpression of MARCKS mRNA is closely related to the occurrence of ischemic damage in hippocampus.
