目的观察青藤碱对NZB/W F1代狼疮鼠外周血中IL-10、TNF-α、抗dsDNA抗体表达的影响。方法32只NZB/W F1代狼疮鼠分为4组:对照组、青藤碱40、120、360 mg/(kg.d)剂量组。分别采用ELISA法和Real-Time RT-PCR法检测NZB/W F1代狼疮鼠外周血中IL-10、TNF-α、抗dsDNA抗体蛋白含量和IL-10、TNF-αmRNA表达。结果3种剂量青藤碱干预3周后,NZB/W F1代狼疮鼠外周血血清中IL-10、TNF-α、抗dsDNA抗体蛋白含量和IL-10、TNF-αmRNA表达均较对照组明显降低(P<0.05,P<0.01)。结论青藤碱在一定程度上能减轻NZB/W F1代狼疮鼠的免疫学异常及炎症反应。
Objective To observe the effect of sinomenine on the expression of IL-10, TNF-α and anti-dsDNA antibody in the peripheral blood of NZB / W F1 wistar rats. Methods Twenty-two New Zealand white rabbits were divided into 4 groups: control group, sinomenine 40,120,360 mg / (kg.d) dose group. The levels of IL-10, TNF-α and anti-dsDNA antibody and the expressions of IL-10 and TNF-α mRNA in the peripheral blood of NZB / W F1 generation were measured by ELISA and Real-Time RT- Results The serum levels of IL-10, TNF-α, anti-dsDNA and the expressions of IL-10 and TNF-α mRNA in the serum of NZB / W F1 generation were significantly higher than those of the control group after 3 doses of sinomenine Decreased (P <0.05, P <0.01). Sinomenine could alleviate the immunological abnormalities and inflammation of NZB / W F1 generation.
