目的:观察百合逍遥散对抑郁模型大鼠行为学及海马5-羟色胺(5-hydroxytryptamine,5-HT)、去甲肾上腺素(norepinephrine,NE)水平的影响,探讨百合逍遥散对抑郁症的干预作用,并观察其与氟西汀是否具有协同抗抑郁作用。方法:60只雌性成年SD大鼠随机分为空白组、模型组、氟西汀组、百合逍遥散组及百合逍遥散联合氟西汀组(中西医结合组),各12只。除空白组外,各组均采用CUMS法造模28 d。造模成功后,氟西汀组给予1.8 mg·(kg·d)-1氟西汀,百合逍遥散组给予12.8 g·(kg·d)-1百合逍遥散,百合逍遥散联合氟西汀组予1.8 mg·(kg·d)-1氟西汀及12.8 g·(kg·d)-1百合逍遥散,空白组及模型组予等量生理盐水,各组液体量均按20 m L·(kg·d)-1计算。共给药28 d。实验过程中观察大鼠体质量变化及行为学改变,并于实验后测定大鼠海马5-HT、NE水平。结果:与模型组比较,给药2周后,3个给药组大鼠体质量明显增长(P<0.01),中西结合组大鼠水平和垂直运动明显增多(P<0.05),总液体消耗明显增加(P<0.01),糖水偏爱率明显上升(P<0.05);给药4周后,同上,并且体质量变化:中西结合组>中药组>西药组,水平运动增加:中西结合组>中药组>西药组,中西结合组大鼠糖水偏爱百分比显著高于另两组(P<0.05);3个给药组大鼠与模型组的5-HT及NE水平具有统计学差异(P<0.01),并且3个给药组海马5-HT及NE水平的趋势均为:中西结合组﹥西药组﹥中药组。结论:百合逍遥散能上调抑郁模型大鼠海马5-HT及NE水平,与氟西汀联用可增强疗效,这可能是百合逍遥散抗抑郁作用的机制之一。

Results: Compared with the model group, the body weight of rats in the three administration groups increased significantly (P <0.01) after 2 weeks of administration, while the level of horizontal and vertical movement in the combination group increased significantly (P <0.05). Conclusion: Lily Xiaoyaosan can up-regulate the levels of 5-HT and NE in the hippocampus of the depression model rats. Combination therapy with fluoxetine can enhance the therapeutic effect, which may be one of the mechanisms of lily Xiaoyaosan anti-depressant effect.
