百合逍遥散对抑郁模型大鼠行为学及海马5-HT、NE的影响

目的:观察百合逍遥散对抑郁模型大鼠行为学及海马5-羟色胺(5-hydroxytryptamine,5-HT)、去甲肾上腺素(norepinephrine,NE)水平的影响,探讨百合逍遥散对抑郁症的干预作用,并观察其与氟西汀是否具有协同抗抑郁作用。方法:60只雌性成年SD大鼠随机分为空白组、模型组、氟西汀组、百合逍遥散组及百合逍遥散联合氟西汀组(中西医结合组),各12只。除空白组外,各组均采用CUMS法造模28 d。造模成功后,氟西汀组给予1.8 mg·(kg·d)-1氟西汀,百合逍遥散组给予12.8 g·(kg·d)-1百合逍遥散,百合逍遥散联合氟西汀组予1.8 mg·(kg·d)-1氟西汀及12.8 g·(kg·d)-1百合逍遥散,空白组及模型组予等量生理盐水,各组液体量均按20 m L·(kg·d)-1计算。共给药28 d。实验过程中观察大鼠体质量变化及行为学改变,并于实验后测定大鼠海马5-HT、NE水平。结果:与模型组比较,给药2周后,3个给药组大鼠体质量明显增长(P<0.01),中西结合组大鼠水平和垂直运动明显增多(P<0.05),总液体消耗明显增加(P<0.01),糖水偏爱率明显上升(P<0.05);给药4周后,同上,并且体质量变化:中西结合组>中药组>西药组,水平运动增加:中西结合组>中药组>西药组,中西结合组大鼠糖水偏爱百分比显著高于另两组(P<0.05);3个给药组大鼠与模型组的5-HT及NE水平具有统计学差异(P<0.01),并且3个给药组海马5-HT及NE水平的趋势均为:中西结合组﹥西药组﹥中药组。结论:百合逍遥散能上调抑郁模型大鼠海马5-HT及NE水平,与氟西汀联用可增强疗效,这可能是百合逍遥散抗抑郁作用的机制之一。 Objective: To observe the effect of Lily Xiaoyao powder on the behavior and the level of 5-hydroxytryptamine (5-HT) and norepinephrine (NE) in hippocampus of rats with depression and to explore the intervention of lily Xiaoyaosan on depression Role, and observe whether it has a synergistic antidepressant effect with fluoxetine. Methods: Sixty female adult Sprague-Dawley rats were randomly divided into blank group, model group, fluoxetine group, Lily Xiaoyao powder group and Lily Xiaoyaosan combined fluoxetine group (Integrative Medicine Group), 12 in each. Except the blank group, CUMS method was used in each group for 28 days. After the model was established, the fluoxetine group was given 1.8 mg · kg -1 dl-fluoxetine, and the Lily Xiaoyao powder group was given 12.8 g · kg -1 d -1 Xiaoyao powder, and the lily Xiaoyao powder combined with fluoxetine The rats in the blank group and the model group were given the same amount of normal saline, and the liquid volume of each group was calculated as 20 m L (mg · kg -1) -1 fluoxetine and 12.8 g · kg -1 d -1 Xiaoyao San · (Kg · d) -1. A total of 28 d administration. During the experiment, we observed the change of body weight and behavior in rats, and measured the level of 5-HT and NE in rat hippocampus after the experiment. Results: Compared with the model group, the body weight of rats in the three administration groups increased significantly (P <0.01) after 2 weeks of administration, while the level of horizontal and vertical movement in the combination group increased significantly (P <0.05). And the trend of 5-HT and NE levels in hippocampus of three administration groups were: Chinese-Western combination group> Western medicine group> Chinese herb group. Conclusion: Lily Xiaoyaosan can up-regulate the levels of 5-HT and NE in the hippocampus of the depression model rats. Combination therapy with fluoxetine can enhance the therapeutic effect, which may be one of the mechanisms of lily Xiaoyaosan anti-depressant effect.
