目的研究DEPDC7基因在正常肝细胞、肝癌细胞及其他肿瘤细胞中的表达情况。方法常规培养正常肝细胞HL-7702,肝癌细胞SMMC7721、HepG2、Huh-7,其他肿瘤细胞ZR-7530、MDA-MB-231、SGC7901、SW480。用MTT检测肝癌细胞转染前后的增殖情况,用RT-PCR方法在转录水平检测DEPDC7基因的表达,用Western blot方法从蛋白质水平检测DEPDC7蛋白的表达。结果 3株肝癌细胞生长增殖速度差异有统计学意义(P<0.05)。肝癌细胞DEPDC7基因在mRNA和蛋白质表达水平比正常肝细胞降低(P<0.05),而其他肿瘤细胞DEPDC7基因表达差异无统计学意义(P>0.05)。结论 DEPDC7基因在正常肝细胞中选择性高表达,而在肝癌细胞株中呈低表达。
Objective To study the expression of DEPDC7 gene in normal hepatocytes, hepatoma cells and other tumor cells. Methods The normal hepatocytes HL-7702, hepatoma cells SMMC7721, HepG2, Huh-7 and other tumor cells ZR-7530, MDA-MB-231, SGC7901 and SW480 were routinely cultured. The proliferation of hepatoma cells was detected by MTT assay. The expression of DEPDC7 gene was detected by RT-PCR at transcriptional level. The expression of DEPDC7 protein was detected by Western blot. Results The proliferation rate of three hepatoma cells was significantly different (P <0.05). The expression of DEPDC7 mRNA and protein in hepatocellular carcinoma cells was lower than that in normal hepatocytes (P <0.05), while there was no significant difference in DEPDC7 gene expression between other tumor cells (P> 0.05). Conclusion DEPDC7 gene is highly expressed in normal liver cells but low in hepatocellular carcinoma cell lines.
