目的初步探讨橡胶防老剂4020和4010NA对人胚肺成纤维细胞(HELF)的毒性差异和剂量-反应趋势。方法橡胶防老剂4020和4010NA以终浓度3、6、9、12μg/ml处理HELF细胞24 h后,应用MTT(噻唑蓝)比色试验观察细胞增殖抑制率。结果橡胶防老剂4020在不同浓度下对HELF细胞的增殖抑制率与4010NA相比差异有统计学意义(P<0.01),且两种都表现出随着剂量增大而细胞毒性的作用增强。结论橡胶防老剂4020比4010 NA毒性低,应在我国轮胎制造行业积极推广应用。
Objective To investigate the toxicity differences and dose-response trends of rubber antioxidant 4020 and 4010NA on human embryonic lung fibroblasts (HELF). Methods HELF cells were treated with 4020 and 4010NA at the final concentrations of 3, 6, 9 and 12 μg / ml for 24 h. MTT (thiazolyl blue) colorimetric assay was used to observe the cell proliferation inhibition rate. Results The inhibitory rate of rubber antioxidant 4020 against HELF cells at different concentrations was statistically different from that at 4010NA (P <0.01), and both of them showed an increase of cytotoxicity with increasing dose. Conclusion The rubber antioxidant 4020 is less toxic than 4010 NA and should be actively promoted and applied in the tire manufacturing industry in China.
