目的 探讨乳腺癌组织 p2 1WAF1基因多态性及其临床意义。 方法 采用聚合酶链反应单链构象多态性技术 (PCR SSCP)法检测 10 0例乳腺癌组织 p2 1WAF1基因的结构变异 ,同时应用免疫组织化学SP法检测乳腺癌组织p2 1WAF1的蛋白表达。结果 5 % ( 2 /4 0 )的对照组组织出现多态性 ,18% ( 18/10 0 )的乳腺癌组织出现两种多态性 ,两组间差异有显著性 ( χ2 =30 .94,P <0 .0 5 ) ;p2 1WAF1基因多态性与蛋白表达 ( r =0 .5 76,P <0 .0 1)和组织学分级 (r =0 .360 ,P <0 .0 1)呈正相关。结论 p2 1WAF1基因多态性在乳腺癌组织中明显增加 ;p2 1WAF1基因多态性可能与促进细胞增殖和增强对癌的易感性有关。
Objective To investigate the polymorphism of p2 1WAF1 gene in breast cancer and its clinical significance. Methods Polymerase chain reaction single strand conformation polymorphism (PCR-SSCP) was used to detect the structural variation of p2 1WAF1 gene in 100 breast cancer tissues. The expression of p2 1WAF1 protein in breast cancer tissues was detected by SP immunohistochemistry. Results The polymorphism was found in 5% (2/40%) of the control group, and there were two polymorphisms in 18% (18/10 0) of the breast cancer tissues. There was a significant difference between the two groups (( 2 =30.94). , P <0. 0 5); P2 1WAF1 gene polymorphism and protein expression (r = 0.576, P <0. 01) and histological grade (r = 0. 360, P <0. ) Positive correlation. Conclusion The polymorphism of p2 1WAF1 gene is significantly increased in breast cancer. The polymorphism of p2 1WAF1 gene may be related to the promotion of cell proliferation and enhanced susceptibility to cancer.
