吉林省为孢子丝菌病高发区,为掌握吉林省自然环境中孢子丝菌的分布状态,我们深入该病高发区通榆、农安及低发区图们等地.采集腐烂芦苇、玉米秸、腐木及土壤.从中分离孢子丝菌.分离菌株再作传代培养,观察菌落形态、颜色、玻片培养镜下生长形态及双相试验等,鉴定菌种,共分离出22株申克孢子丝菌.从通榆的60份陈旧芦苇中分离出3株申克孢子丝菌,分离率为5%.从农安洼中高农场腐烂芦苇(20份)中分离出9株申克孢子丝菌,分离率45%;从腐烂玉米秸(20份)中分离出2株,分离率10%;从土壤(20份)中分离出3株,分离率为15%.从低发区图们江流域珲春、石砚等地苇塘内腐烂芦苇(40份)中分离出3株申克孢子丝菌.分离率7.5%;从土壤(20份)中分离出1株,分离率5%;从20份腐木中分离出1株,分离率5%.以上分离所得22株孢子丝菌.初代菌落均为乳白色酵母样,1~2周后由中心部开始逐渐呈褐黑色并见皱褶.部分菌落表面最终呈褐色粉末状,部分菌落最终呈淡黄褐色,亦见皱褶.全部菌株玻片培养.镜下均见有分枝分隔细长菌丝,并见沿菌丝两侧排列的分生孢子及在分生孢子柄顶端呈花弁状排列的分生孢子.分生孢子柄与菌丝分枝角度为90°或小于90°,分生孢子为圆形或椭圆形单细胞性孢子.双相试验结果.菌丝型菌株转为酵母型,分生孢子大小在2μm×3μm~
