建立了简易的制备~(32)P-酪蛋白和测定核内磷蛋白磷酸酶的方法。测定了Hep.A腹水肝癌染色质结合的蛋白激酶和核内磷蛋白磷酸酶活性。Hep.A肝癌染色质结合的蛋白激酶比活性大于正常肝,但核内磷蛋白磷酸酶的比活性反低于正常。这是引起Hep.A肝癌染色质总磷酸化修饰程度增加,非组蛋白含磷总量增加的重要原因。Hep.A肝癌染色质磷酸化作用的增长低于非组蛋白量的增加,可能是引起单位重量磷酸化非组蛋白所含磷酸基团低于正常肝的原因之一。
A simple method for the preparation of ~ (32) P-casein and determination of the nuclear phosphoprotein phosphatase was established. The chromatin-bound protein kinase and the nuclear phosphoprotein phosphatase activity of Hep.A ascites hepatocellular carcinoma were determined. Hep.A hepatocellular carcinoma chromatin-associated protein kinase specific activity than normal liver, but the specific activity of the nuclear phosphoprotein phosphatase anti-less than normal. This is caused by Hep.A liver cancer chromatin total phosphorylation degree of increase, an increase of non-histone phosphorus an important reason. Hep.A HCC chromatin phosphorylation increased less than non-histone increase, may be caused by per unit weight of phosphorylated non-histone phosphate group is lower than normal liver one of the reasons.
