目的 探索一种新的脑胶质瘤生长抑制因子Glob-N的分离鉴定方法。方法 人脑中性鞘糖脂经过加压柱层析,梯度甲醇/氯仿溶液洗脱。对所得到的产物行1H-NMR和13C-NMR测定。结果 含甲醇60%的甲醇/氯仿溶液的洗脱物为纯化Glob-N,层析展开后呈单一清晰条带,Rf值0.45。初步结构分析表明人脑Glob-N含N-乙酰半乳糖,共3个糖基,其神经酰胺部分共有33个碳原子。结论 柱层析分离中性鞘糖脂单一组分效果好,纯度高;人脑Glob结构中含3个糖基。
Objective To explore a new method for the isolation and identification of glioma growth inhibitory factor Glob-N. Methods Human brain neutral glycosphingolipid was separated by pressurized column chromatography and eluted with gradient methanol / chloroform solution. The obtained product was subjected to1H-NMR and13C-NMR measurements. Results The eluate containing 60% methanol in methanol / chloroform solution was purified Glob-N. The chromatogram showed a single clear band after it was developed. The Rf value was 0.45. Preliminary structural analysis showed that human brain Glob-N contains N-acetylgalactose, a total of three glycosyl groups, the ceramide portion of a total of 33 carbon atoms. Conclusion Column chromatography separation of neutral glycosphingolipid single component effective, high purity; human brain Glob structure containing three sugar.
