肠胃清含药血清对人结肠癌HCT116/L-OHP耐药细胞凋亡作用的影响

目的探讨中药复方肠胃清(CWQ)逆转人结肠癌HCT116/L-OHP细胞耐药的分子机制。方法提取肠胃清含药血清,设置不同浓度,采用MTT法确定肠胃清的最大无毒剂量和逆转指数(RI);采用彗星实验法检测肠胃清联合奥沙利铂(L-OHP)对耐药细胞DNA损伤的影响,流式细胞术观察肠胃清联合奥沙利铂(L-OHP)对耐药细胞凋亡的影响。结果肠胃清含药血清可以逆转人结肠癌HCT116/L-OHP耐药细胞对L-OHP的耐药性,逆转指数(RI)为2.63倍。肠胃清联合L-OHP能促进耐药细胞发生DNA损伤(P<0.001)及凋亡(P<0.01)。结论肠胃清能够逆转人结肠癌铂类化疗耐药,其机制与促进肿瘤细胞DNA损伤、诱导凋亡有关。 Objective To investigate the molecular mechanism of traditional Chinese medicine compound weiweiqing (CWQ) on reversing the drug resistance of human colon cancer HCT116 / L-OHP cells. Methods The serum containing serum of Changweiqing was extracted and set at different concentrations. The maximum non-toxic dose and reversal index (RI) of gastrointestinal juice were determined by MTT method. The effect of L-OHP combined with L-OHP on drug resistance Cell DNA damage, flow cytometry observed gastro-qingqing combined oxaliplatin (L-OHP) on apoptosis of drug-resistant cells. Results The GXG serum could reverse the resistance of human colon cancer HCT116 / L-OHP cells to L-OHP. The reversal index (RI) was 2.63 times. Gastrointestinal combined with L-OHP can promote DNA damage (P <0.001) and apoptosis in drug-resistant cells (P <0.01). Conclusion Gengweiqing can reverse the drug resistance of human colon cancer with platinum-based chemotherapy. Its mechanism is related to DNA damage and induction of apoptosis in tumor cells.
