细胞能量异常是肿瘤细胞的十大特征之一。快速增殖的癌细胞生长所需要的能量主要是由糖酵解而非氧化磷酸化提供,癌细胞这种特殊的糖代谢被称为Warburg效应。近年来肿瘤的靶向治疗成为研究的热点,异常的糖酵解、脂代谢和氨基酸代谢的过程涉及到大量的限速酶和关键酶,这些都是潜在的药物治疗靶点。本文仅围绕着调控癌细胞糖酵解进程中直接调控的关键酶和限速酶,脂代谢过程中脂肪酸合成酶和谷氨酰胺代谢过程中的谷氨酰胺酶进行分析和综述。
Abnormal cell energy is one of the top ten features of tumor cells. The energy required for the rapidly proliferating cancer cells to grow is provided primarily by glycolysis rather than oxidative phosphorylation, and this particular glycometabolism of cancer cells is called the Warburg effect. In recent years, targeted therapy has become a hot research topic. Abnormal glycolysis, lipid metabolism and amino acid metabolism involve a large number of rate-limiting enzymes and key enzymes, all of which are potential drug targets. This article focuses on the regulation and control of cancer cells in the process of glycolysis directly regulates the key enzyme and rate-limiting enzyme, lipid metabolism during fatty acid synthase and glutamine metabolism in the process of glutaminase analysis and review.
