目的优化中药水提液膜分离前的预处理方式。方法以黄连解毒汤水提液中4种共性高分子物质和小分子药效物质(小檗碱、栀子苷)组成的综合模拟体系为研究对象,通过不同预处理方式调节溶液环境,分别考察不同溶液环境对膜通量、共性高分子物质去除率、小分子药效物质透过率等指标的影响。结果调节pH值对膜通量的改善较为明显,共性高分子物质截留率较高;调节pH值和离子强度(加入0.05 mol/L NaCl)时小分子药效物质透过率相对理想。研究发现,膜堵塞阻力是膜污染的主要因素。结论综合各项指标,黄连解毒汤以调节离子强度和调节pH值为6.5时的预处理效果较为理想;对于中药水提液的膜分离前预处理具有一定的参考意义。
Objective To optimize the pretreatment method of traditional Chinese medicine aqueous extract before membrane separation. Methods A comprehensive simulation system composed of four kinds of common macromolecule substances and small molecule pharmacological substances (berberine and geniposide) in the water extract of berberine and toxin decoction was used as research object. Different pretreatment methods were used to adjust the solution environment, Effect of solution environment on membrane flux, common macromolecule material removal rate, small molecule drug substance permeability and other indexes. Results The adjustment of pH value showed a significant improvement on membrane flux. The rejection rate of common macromolecule material was higher. The transmittance of small molecule drug substance was relatively good when the pH and ionic strength were adjusted (0.05 mol / L NaCl addition). The study found that membrane clogging resistance is the main factor of membrane fouling. Conclusion According to the various indexes, the pretreatment effect of berberine detoxification decoction to adjust the ionic strength and adjust the pH to 6.5 is ideal. It is of certain reference significance for the pretreatment before the membrane separation of traditional Chinese medicine aqueous extract.
