利用单因子试验和响应面法优化了影响灵芝菌丝体灵芝酸的提取过程。基于3因素3水平的中心组合设计,得到了描述胞内灵芝酸提取得率与操作参数之间的二次响应面模型。在乙醇浓度为93.6%(v/v)、提取温度79.1℃、提取液固比42.2∶1、提取次数2次、每次提取时间2 h的条件下,最大灵芝酸理论提取得率为28.36 mg胞内灵芝酸每克干菌丝。模型优化条件下的实际灵芝酸提取得率(28.72 mg/g干菌丝)与理论灵芝酸提取得率(28.36 mg/g干菌丝)相符。
The single factor experiment and response surface method were used to optimize the extraction process of Ganoderma lucidum which affects the mycelia of Ganoderma lucidum. Based on the central combination design of 3 factors and 3 levels, a quadratic response surface model describing the extraction yield and operating parameters of intracellular ganoderic acid was obtained. At the ethanol concentration of 93.6% (v/v), the extraction temperature of 79.1°C, the extract solids ratio of 42.2∶1, the extraction times of 2 times, and the extraction time of 2 h, the maximum theoretical extraction yield of ganoderic acid was 28.36 mg. Intracellular ganoderic acid per gram of dry mycelium. The actual ganoderic acid extraction yield (28.72 mg/g dry hyphae) under the optimized conditions was consistent with the theoretical Ganoderma acid extraction yield (28.36 mg/g dry hyphae).
