目的探讨步长脑心通胶囊对急性心肌梗死(AMI)兔的心功能及梗死面积的作用。方法将42只新西兰兔随机分为假手术组、梗死对照组、药物组,每组14只。采用手术结扎左冠状动脉前降支(LAD)建立AMI兔模型,假手术组反穿线不结扎,各组均造模成功6只。假手术组、梗死对照组给予常规喂养,药物组在此基础上加入步长脑心通胶囊每次0.4g,每日3次,实验周期为3周,分别于术后2天、21天行心脏超声检查,检测左室舒张末期内径(LVDd)、左室收缩末期内径(LVDs)、左室射血分数(LVEF),21天后处死家兔行心肌梗死面积测定。结果术后梗死对照组及药物组即刻可见左室前壁及心尖部颜色变暗,搏动减弱,心电图显示两个以上导联出现ST段持续弓背向上抬高并与T波融合;术后21天药物组与梗死对照组比较,LVEF增高(P<0.05),而梗死面积明显降低(P<0.05);与本组术后2天比较,药物组术后21天LVEF增加(P<0.05)。结论步长脑心通胶囊能够显著降低AMI兔术后21天心肌梗死面积,增加LVEF,改善心脏收缩功能。
Objective To investigate the effect of Buchang Naoxintong Capsule on cardiac function and infarct size in rabbits with acute myocardial infarction (AMI). Methods Forty-two New Zealand white rabbits were randomly divided into sham operation group, infarction control group and drug group, with 14 rats in each group. Surgical ligation of the left anterior descending coronary artery (LAD) to establish the AMI rabbit model, sham-operated group anti-threading ligation, the success of each model were 6. Sham-operation group and infarction control group were given conventional feeding. On the basis of this, the drug group was given BuXiaoNaoTongTong capsule 0.4g each time, the experimental period was 3 weeks, respectively, at 2 days and 21 days after operation The left ventricular end-diastolic diameter (LVDd), left ventricular end-systolic diameter (LVDs) and left ventricular ejection fraction (LVEF) were measured by echocardiography, and infarction area of rabbits sacrificed 21 days later. Results Immediately after operation, the color and darkening of the anterior wall of the left ventricle and apical part of the infarction control group and the drug group showed weakened beat. Electrocardiogram showed that the ST segment continued to rise upwards and merged with T wave in two or more leads. After operation 21 LVEF increased (P <0.05) and infarct size decreased significantly in the drug group compared with the infarction control group (P <0.05). Compared with the 2 days after operation, the LVEF in the drug group increased 21 days after operation (P <0.05) . Conclusion Bu Tong Naoxintong capsule can significantly reduce myocardial infarction area 21 days after AMI rabbits, increase LVEF, improve cardiac contractility.
