观察小檗胺及其衍生物(B2、B4、BB、EBB)对培养的恶性黑色素瘤细胞(BowesCell)增殖都有明显的抑制作用,其抑制增殖的IC50值分别为小檗胺(Bo)9.5μmol、B2:3.34μmol、B4:3.04μmol、BB:4.5μmol、EBB:1.5μmol。对3H-TdR掺入和单细胞集落形成(克隆)以及脱氢酶活性同样有明显的抑制作用,其作用特点是经化学修饰后的小梁胺的作用比母体小檗胺强;而4种衍生物中,EBB的抑制效果最强。
Observed that berbamine and its derivatives (B2, B4, BB, EBB) have significant inhibition on the proliferation of cultured melanoma cells (BowesCell), and their IC50 values for inhibiting proliferation are berbamine (Bo) 9 .5 μmol, B2: 3.34 μmol, B4: 3.04 μmol, BB: 4.5 μmol, and EBB: 1.5 μmol. The inhibition of 3H-TdR incorporation and single cell colony formation (cloning) and dehydrogenase activity was also evident. The effect of the chemically modified trabecamine was stronger than that of the parent berbamine; Among the derivatives, EBB has the strongest inhibitory effect.
