目的 探讨C myc基因作为Wnt/Frizzled信号通路靶基因在肾癌生物学行为中的表达及其意义。 方法 采用细胞核蛋白印迹 (Western blot)、免疫组化及免疫细胞化学C myc蛋白半定量分析方法 ,对 32例肾癌组织及肾癌细胞株GRC I、RCC 949进行了研究。 结果 C myc在肾癌组织中阳性表达 ,细胞浆阳性率 90 .6 % (2 9/ 32 )、细胞核 75 .0 % (2 4/ 32 ) ,其表达与肾癌细胞组织分型、临床分期有较大相关性 ;C myc在GRC I中呈强阳性表达。 结论 Wnt/Frizzled信号通路及其靶基因C myc在肾癌的发生发展中具有重要作用 ,通过Wnt/Frizzled信号通路的干预 ,以C myc为靶分子的肿瘤信号转导通路治疗将有重要价值及应用前景
Objective To investigate the expression and significance of C myc gene as a target gene of Wnt / Frizzled signaling pathway in biological behavior of renal cell carcinoma. Methods 32 cases of renal cell carcinoma and renal cell carcinoma cell lines GRC I and RCC 949 were studied by Western blot, immunohistochemistry and semi-quantitative immunocytochemistry Cmyc. Results The positive expression rate of C-myc in renal cell carcinoma was 90.6% (29/32) and 75.0% (24/32) in renal cell carcinoma. The expression of C-myc was correlated with the histological type, There was a strong correlation between Cmyc and GRC I expression. Conclusion The Wnt / Frizzled signaling pathway and its target gene Cmyc play an important role in the development of renal cell carcinoma. The intervention of Wnt / Frizzled signaling pathway is of great value in the treatment of tumor signal transduction pathway targeting C-myc and Application prospects
