目的探讨波动性高糖下血红素加氧酶1(HO-1)对INS-1细胞氧化应激的影响。方法以体外培养的INS-1细胞为研究对象,分为正常对照组、持续性高糖组、波动性高糖组、钴原卟啉(CoPP)+波动性高糖组、锌原卟啉(ZnPP)+波动性高糖组。各组均干预72h,Western blot检测HO-1蛋白表达水平,流式细胞仪测定活性氧簇(ROS)水平,四氮唑蓝(MTT)法测定细胞活力。结果波动性高糖组HO-1蛋白表达及ROS水平均显著高于持续性高糖组(P均<0.01),波动性高糖组细胞活力显著低于持续性高糖组(P<0.05);与波动性高糖组相比,CoPP+波动性高糖组HO-1表达水平及细胞活力升高、ROS水平下降(P均<0.05),而ZnPP+波动性高糖组呈现相反的变化趋势。结论波动性高糖较持续性高糖引起INS-1细胞产生更严重的氧化应激,在波动性高糖条件下,HO-1可能通过抗氧化应激对INS-1细胞发挥保护效应。
Objective To investigate the effect of heme oxygenase 1 (HO-1) on oxidative stress in INS-1 cells under fluctuating high glucose. Methods INS-1 cells cultured in vitro were divided into normal control group, continuous high glucose group, high glucose group, CoPP + high glucose group, zinc protoporphyrin ZnPP) + fluctuating high glucose group. Each group was intervened for 72 hours, the expression of HO-1 protein was detected by Western blot, the level of reactive oxygen species (ROS) was determined by flow cytometry, and the cell viability was measured by MTT assay. Results The HO-1 protein expression and ROS level in high glucose group were significantly higher than those in high glucose group (all P <0.01). The cell viability in high glucose group was significantly lower than that in high glucose group (P <0.05) (P <0.05). Compared with high glucose group, the expression of HO-1 and cell viability were increased and ROS level was decreased in CoPP + high glucose group (P <0.05). CONCLUSION: High-glucose and high-glucose may cause more severe oxidative stress in INS-1 cells. Under fluctuating high glucose conditions, HO-1 may exert a protective effect on INS-1 cells through anti-oxidative stress.
