目的观察龟鹿二仙胶及其拆方对豚鼠膝关节软骨功能基因表达的影响,探讨龟鹿二仙胶的配伍机理。方法取3月龄豚鼠膝关节软骨建立软骨细胞体外培养体系,采用免疫组化染色、RT-PCR、Elisa方法检测酸性GAG、Ⅱ型胶原、Bax、Bcl-2、iNOs的表达。结果龟鹿参组能显著促进软骨细胞分泌酸性GAG(P<0.05),促进Ⅱ型胶原合成(P<0.05);龟鹿杞组能显著抑制Bax的表达、促进Bcl-2表达,抑制软骨细胞分泌iNOs(P<0.05),二组的作用与龟鹿二仙胶组相同。结论人参可助龟甲胶、鹿角胶促进软骨细胞分泌酸性GAG和Ⅱ型胶原,枸杞子可助龟甲胶、鹿角胶延缓软骨细胞凋亡,抑制软骨细胞分泌iNOs。
Objective To observe the effect of Guilu Erxian and its decomposed formulas on the expression of functional genes in guinea pig articular cartilage and to explore the compatibility mechanism of Guilu Erxian gum. Methods Chondrocytes were cultured in 3-month-old guinea pig articular cartilage in vitro. The expressions of acidic GAG, type Ⅱ collagen, Bax, Bcl-2 and iNOs were detected by immunohistochemistry, RT-PCR and Elisa methods. Results Guifu group could significantly promote the secretion of acid GAG (P <0.05) and promote the synthesis of type Ⅱ collagen (P <0.05). Guikuqi group could significantly inhibit the expression of Bax, promote the expression of Bcl-2 and inhibit the proliferation of chondrocytes Secretion of iNOs (P <0.05), the role of two groups and Guilu Erxian glue the same group. CONCLUSION Ginseng can help tortoise shell glue and antler glue to promote the secretion of acid GAG and type Ⅱ collagen in chondrocytes. Medlar can help tortoise shell glue and antler glue to retard chondrocyte apoptosis and inhibit the secretion of iNOs by chondrocytes.
