目的通过建立人胃癌裸鼠腹腔种植瘤模型,探讨CO2气腹对胃癌组织中乙酰肝素酶(HPA)、血管内皮生长因子(VEGF)表达的影响及意义。方法27只裸鼠分3组:对照组、5mmHgCO2气腹组及8mmHgCO2气腹组,每组9只。人胃癌细胞SGC-7901裸鼠腹腔注射后建立CO2气腹胃癌细胞裸鼠腹腔种植模型,气腹持续1h。处理后4周处死解剖裸鼠,记录腹腔各脏器、穿刺口和腹壁切口成瘤情况和肿瘤数,测量肿瘤最大直径、总质量,并检测胃癌种植瘤组织中HPAmRNA和蛋白表达及VEGF蛋白的表达。结果各组裸鼠腹腔种植瘤结节数、平均最大直径、质量相比无统计学差异(P>0.05)。各组裸鼠胃癌组织中HPA和VEGF的表达水平相比无统计学差异(P>0.05)。结论5mmHg和8mmHgCO2气腹对胃癌细胞裸鼠腹腔内增殖和侵袭转移无明显影响,也未明显促进种植瘤组织中HPA和VEGF的表达。
Objective To establish a model of peritoneal xenografts of human gastric cancer in nude mice and investigate the effect and significance of CO2 pneumoperitoneum on the expression of heparanase (HPA) and vascular endothelial growth factor (VEGF) in gastric cancer. Methods Twenty-seven nude mice were divided into three groups: control group, pneumoperitoneum of 5mmHgCO2 and pneumoperitoneum of 8mmHgCO2, with 9 in each group. Gastric cancer cells SGC-7901 nude mice after intraperitoneal injection of CO2 pneumoperitoneum to establish a model of intraperitoneal implantation of pneumoperitoneum, pneumoperitoneum for 1h. The nude mice were sacrificed 4 weeks after the operation and the tumor size and the number of tumor in the abdominal organs, puncture and abdominal incision were recorded. The maximum diameter and the total mass of tumor were measured. The expression of HPA mRNA and protein and VEGF protein expression. Results There was no significant difference in the number of nodules, average maximum diameter and mass of nude mice in each group (P> 0.05). There was no significant difference in the expression of HPA and VEGF in each group of gastric cancer (P> 0.05). Conclusion Pneumoperitoneum of 5mmHg and 8mmHgCO2 had no significant effect on the proliferation, invasion and metastasis of gastric cancer cells in nude mice, and did not promote the expression of HPA and VEGF in implanted tumor.
