目的:测定急性出血坏死性胰腺炎(AHNP)大鼠血浆及胰腺组织内皮素(ET)含量变化,探讨其在发病机理中的作用。方法:5%牛磺胆酸钠溶液胆胰管注入,诱发大鼠AHNP。动态测定血浆及胰腺组织内皮素(ET)、胰腺血流量(PBF)、肌醇三磷酸(IP3)和胰腺细胞[Ca2+]i的变化,胰腺组织病理评分(PM)定量分析。结果:AHNP大鼠血浆及胰组织ET含量早期显著增加,血浆ET初始升高滞后于胰组织ET,两者间及两者与PBF、IP3、[Ca2+]i、PM呈显著相关性,但胰组织ET相关更密切。结论:胰组织ET可能介导了胰腺早期缺血,血浆ET进一步加重AHNP大鼠的病理生理改变。
Objective: To determine the content of endothelin (ET) in plasma and pancreas of acute hemorrhagic necrotizing pancreatitis (AHNP) rats and to explore its role in pathogenesis. Methods: 5% sodium taurocholate solution was injected into the biliary and pancreatic ducts to induce AHNP in rats. The levels of endothelin (ET), pancreatic blood flow (PBF), inositol triphosphate (IP3) and pancreatic cells [Ca2 +] i in plasma and pancreatic tissue were measured dynamically. Quantitative analysis of pancreatic histopathological score (PM) Results: The content of ET in plasma and pancreatic tissue of AHNP rats increased significantly at the early stage, and the initial ET of plasma ET lapsed later than that of pancreatic tissue ET. There was a significant correlation between ET and PBF, IP3, [Ca2 +] i and PM, Tissue ET is more closely related. Conclusion: ET in pancreatic tissue may mediate the early ischemia of pancreas and the pathophysiological changes of AHNP rats are further aggravated by ET.
