目的:观察ω-3多不饱和脂肪酸(ω-3PUFAs)对MGC803低分化胃癌细胞系凋亡的影响,并对其机制进行探讨。方法:采用四甲基偶氮唑蓝(MTT)观察细胞增殖率、DNA电泳和流式细胞术(FCM)检测细胞凋亡情况、蛋白质印迹法检测凋亡蛋白PARP-1和AIF的表达。结果:二十碳五烯酸(EPA)和二十二碳六烯酸(DHA)通过诱导细胞凋亡显著抑制胃癌细胞的生长,呈时间和剂量依赖关系。DHA促进细胞凋亡的作用强于EPA。40μg/mL EPA和DHA作用MGC803细胞24 h后,凋亡相关蛋白PARP-1和AIF表达均较对照组显著增加。结论:ω-3 PUFAs通过诱导细胞凋亡抑制胃癌细胞的增殖,其机制可能通过PARP-1/AIF凋亡通路实现。
Objective: To observe the effect of ω-3 polyunsaturated fatty acids (ω-3 PUFAs) on the apoptosis of MGC803 poorly-differentiated gastric cancer cell lines and to explore its mechanism. Methods: Cell proliferation was observed by MTT assay. Cell apoptosis was detected by DNA electrophoresis and flow cytometry (FCM). The expressions of PARP-1 and AIF were detected by Western blotting. Results: Eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) significantly inhibited the growth of gastric cancer cells by inducing apoptosis in a time-and dose-dependent manner. DHA can promote apoptosis better than EPA. After treated with 40μg / mL EPA and DHA for 24 h, the expressions of PARP-1 and AIF in MGC803 cells increased significantly compared with the control group. CONCLUSION: Omega-3 PUFAs can inhibit the proliferation of gastric cancer cells by inducing apoptosis. The mechanism may be through the PARP-1 / AIF apoptosis pathway.
