用脉冲场凝胶电泳分离回收覆盖脆性X位点的酵母人工染色体YAC209G4中475kb片段,经MboⅠ完全酶解为平均长度400bp的片段,与pBSⅡKS质粒载体重组,共得到3500个重组作,筛选出60个含单拷贝序列插入片段的克隆。经序列测定,并与基因数据库分析比较,其中28个序列被基因数据库接受为新的序列标签位点,GenBank收录号为U26560-U26587。根据其中3个新序列标签的核普酸顺序设计PCR引物,在人基因组DNA中扩增出相应的特异片段。
A 475kb fragment of yeast artificial chromosome YAC209G4 covering the fragile X locus was isolated and purified by pulsed-field gel electrophoresis. The fragment was digested to the average length of 400bp by MboⅠ and recombined with pBSⅡKS plasmid vector to obtain 3,500 recombinants. Clone with single copy insertions. After sequenced and compared with the gene database, 28 of them were accepted as new sequence tagging sites by GenBank. The GenBank accession number was U26560-U26587. PCR primers were designed based on the nucleotide sequences of three new sequence tags, and corresponding specific fragments were amplified from human genomic DNA.
