[目的]了解变蓝菌素对口腔鳞癌及口腔正常粘膜内源性脂肪酸合成活性的抑制特点。[方法]切取口腔鳞癌患者新鲜手术标本的癌组织及正常口腔粘膜组织块 ,将组织块剪成细块 ,分变蓝菌素处理组与对照组。用[U- 14C]乙酸钠示踪 ,液体闪烁计数14C的掺入量 ,以显示脂肪酸合成活性。[结果]口腔鳞癌组织块在变蓝菌素处理1h时脂肪酸合成活性约下降19% ,2h下降约64% ,4h下降约87 % ;对照组轻度上升。口腔粘膜在变蓝菌素处理后脂肪酸合成活性略下降 ,至4h时仅下降20% ;对照组略微上升。[结论]变蓝菌素能明显抑制口腔鳞癌组织的内源性脂肪酸合成 ,但对口腔正常粘膜组织作用轻微 ,提示口腔鳞癌脂肪酸合成途径有可能成为研发抗肿瘤治疗药物的靶目标。
[Objective] To understand the inhibitory effect of streptogramin on the activity of endogenous fatty acid synthesis in oral squamous cell carcinoma and oral mucosa. [Method] The cancerous tissue and normal oral mucosa tissue of fresh surgical specimens of patients with oral squamous cell carcinoma were excised, and the tissue pieces were cut into thin pieces and divided into plastid strain and control group. Traced with [U-14C] sodium acetate, the amount of incorporation of 14C was scintillated by liquid scintillation to show fatty acid synthesis activity. [Result] The fatty acid synthesis activity of oral squamous cell carcinoma tissue decreased by 19% at 1 h, decreased by 64% at 2 h, decreased by 87% at 4 h and slightly increased in control group. The activity of fatty acid synthesis in oral mucosa decreased slightly after treatment with cercin, only decreased by 20% at 4h; the control group increased slightly. [Conclusion] Flucystatin can significantly inhibit the synthesis of endogenous fatty acids in oral squamous cell carcinoma tissues, but it has a slight effect on normal oral mucosa tissues, suggesting that the fatty acid synthesis pathway of oral squamous cell carcinoma may become the target of developing anti-tumor therapeutic drugs.
