目的 介绍实验性变态反应性脑脊髓炎 (EAE)的致敏抗原———髓鞘素的一种制备方法。方法 取牛脊髓白质加入蔗糖溶液制成脊髓匀浆 ,在 4℃通过不连续梯度离心 ,得到粗制的髓鞘素 ,进一步去除其中的磷脂、胆固醇和核酸等成分 ,得到纯化的髓鞘素。计算其含量 ,检测其主要成分及相对分子质量 ,并通过对豚鼠进行免疫 ,检测其诱发EAE的活性。结果 所提取的髓鞘素具有与文献报道结果类似的理化特性和强大的EAE诱发活性。结论 本方法提纯的髓鞘素有强的免疫活性 ,易取材 ,操作简便 ,便于推广。
Objective To introduce a method for the preparation of myelin, a sensitizing antigen in experimental allergic encephalomyelitis (EAE). Methods Bovine spinal cord white matter was added into sucrose solution to make spinal cord homogenate. Centrifuged at 4 ℃ by discontinuous gradient centrifugation, the crude myelin was obtained and the phospholipids, cholesterol and nucleic acids were further removed to obtain purified myelin. Calculate the content, detect its main components and relative molecular mass, and through the guinea pigs were immunized to detect the activity of induced EAE. Results The extracted myelin had similar physical and chemical properties and strong EAE-induced activity as reported in the literature. Conclusion The myelin purified by this method has strong immunocompetence and is easy to be taken, easy to operate and easy to popularize.
