目的研究忍冬Lonicera japonica根中的化学成分及其抗炎活性。方法采用反复碱性硅胶柱色谱、Sephadex LH-20柱色谱、薄层色谱、制备液相色谱等方法进行分离纯化,并通过波谱学分析鉴定其化学结构;采用斑马鱼抗炎模型,测定分离所得部分单体成分的抗炎活性。结果从忍冬根95%乙醇提取物中分离得到14个化合物,并鉴定其结构为3,13-dihydroxystemodan-2-one(1)、大黄酚(2)、palmarumycin CP2(3)、β-谷甾醇(4)、豆甾醇(5)、豆甾-4,6,8(14),22-四烯-3-酮(6)、erythrinassinate D(7)、羊毛甾醇(8)、齐墩果酸-3-乙酸酯(9)、原儿茶醛(10)、胡萝卜苷(11)、(E)-3-(3,4-二羟基苯亚甲基)-5-(3,4-二羟基苯)-2(3H)-呋喃酮(12)、lomacarinoside B(13)、(2E,6S)-8-(α-L-arabinopyranosyl-(1″→6′)-β-D-glucopyranosyloxy)-2,6-dimethyloct-2-eno-1,2″-lactone(14)。结论化合物1为新化合物,化合物2、3、6、7~9、13均为首次从该植物中分离得到;化合物9、10在100μg/m L质量浓度下表现出明显的抗炎活性。
Aim To study the chemical constituents and anti-inflammatory activity of Lonicera japonica roots. Methods The compounds were separated and purified by repeated basic silica gel column chromatography, Sephadex LH-20 column chromatography, thin layer chromatography and preparative liquid chromatography, and their chemical structures were identified by spectroscopic analysis. The zebrafish anti-inflammatory model Anti-inflammatory activity of some monomeric components. Results Fourteen compounds were isolated from the 95% ethanol extract of Lonicera japonica root and identified as 3,13-dihydroxystemodan-2-one (1), chrysophanol (2), palmarumycin CP2 (3) (4), stigmasterol (5), stigmasterol-4,6,8 (14), 22-tetraen-3-one (6), erythrinassinate D (7), lanosterol (9), protocatechuic aldehyde (10), daucosterol (11), (E) -3- (3,4-dihydroxybenzylidene) -5- L-arabinopyranosyl- (1 “→ 6 ’) - β-D-glucopyranosyloxy (12) ) -2,6-dimethyloct-2-eno-1,2 ”-lactone (14). Conclusion Compound 1 is a novel compound. Compounds 2,3,6,7 ~ 9,13 were isolated from this plant for the first time. Compound 9,10 showed obvious anti-inflammatory activity at a concentration of 100μg / mL.
