目的探讨十溴联苯醚(decabromodiphenylether,BDE-209)对青春期雄性ICR小鼠的生殖毒性。方法青春期雄性ICR小鼠随机分为6组:对照组(生理盐水),DMSO组,BDE-209处理组(100,200,300,500 mg/kg BW)。小鼠经口染毒7周后处死,称量小鼠睾丸、附睾重量并计算脏器系数。采用放射免疫法检测小鼠血清中睾酮水平,采用ELISA法检测小鼠血清中cAMP水平。结果与对照组相比,DMSO组未见明显生殖损害(均有P>0.05),而BDE-209处理组睾丸重和附睾重、睾丸脏器系数和附睾脏器系数均显著降低(均有P<0.05);与对照组相比,BDE-209处理组小鼠精子数目亦减少(均有P<0.05)。进一步研究发现,BDE-209处理后,小鼠血清cAMP浓度和睾酮浓度低于对照组(均有P<0.05)。结论 BDE-209暴露青春期雄性小鼠导致小鼠睾酮合成降低,精子合成减少,损伤青春期雄性小鼠生殖功能。
Objective To investigate the reproductive toxicity of decabromodiphenylether (BDE-209) to adolescent male ICR mice. Methods Adolescent male ICR mice were randomly divided into 6 groups: control group (saline), DMSO group and BDE-209 treatment group (100, 200, 300 and 500 mg / kg BW). The mice were sacrificed 7 weeks after oral administration of the testicle, the weight of the testis and epididymis were weighed and the organ coefficient was calculated. The level of testosterone in serum of mice was detected by radioimmunoassay. The level of cAMP in serum of mice was detected by ELISA. Results Compared with the control group, there was no significant reproductive impairment in DMSO group (all P> 0.05), while the weight of testis and epididymis, organ coefficient of testis and organ coefficient of epididymis were significantly decreased in BDE-209 group <0.05). Compared with the control group, the number of spermatozoa in BDE-209-treated mice also decreased (all P <0.05). Further study found that BDE-209 treatment, serum cAMP and testosterone concentrations were lower than the control group (all P <0.05). Conclusion Exposure of BDE-209 to adolescent male mice results in decreased testosterone synthesis, reduced sperm synthesis, and impaired reproductive function in adolescent male mice.
