目的:观察损伤血管细胞周期蛋白Β1(CyclinB1)的表达情况,探讨益气活血方对球囊损伤大鼠颈总动脉内膜平滑肌细胞(VSMCs)增殖的干预机制。方法:随机将90只雄性SD大鼠分为模型组,阿司匹林组,益气活血方大、中、小剂量组。球囊损伤大鼠颈总动脉前给予高脂饲料喂养30天,造成高脂血症;然后经右股动脉穿刺球囊扩张损伤大鼠左颈总动脉,造成颈总动脉管腔的狭窄。手术前1天开始给药,每组动物于术后(5d、14d、28d)处死,取损伤颈总动脉,常规固定、包埋、切片,分别做HE染色和免疫组织化学SABC法检测CyclinB1的表达。结果:模型缓慢CyclinB1表达明显增多,用益气活血方干预后,14d和28dCyclinB1表达量与模型组比较差异有显著性(P<0.05)。结论:益气活血方对球囊损伤后动脉血管内膜VSMCs增生有明显的抑制作用,其机制可能与下调细胞周期蛋白B1的表达,抑制血管内膜VSMCs的细胞分裂周期进程有关。
OBJECTIVE: To observe the expression of CyclinB1 and explore the mechanism of Yiqi Huoxue decoction on the proliferation of carotid artery intima-media smooth muscle cells (VSMCs) after balloon injury in rats. Methods: 90 male SD rats were randomly divided into model group, aspirin group, Yiqi Huoxue Fang large, medium and small dose groups. Balloon injury rats before the common carotid artery feeding high-fat feed for 30 days, resulting in hyperlipidemia; and then by the right femoral artery puncture balloon injury rat left common carotid artery, causing the common carotid artery stenosis. The animals were sacrificed on the 1st day before surgery (5d, 14d, 28d). The common carotid arteries were fixed, embedded, and sectioned. HE staining and immunohistochemical SABC method were used to detect the expression of CyclinB1 expression. Results: The expression of CyclinB1 was significantly increased in the model group. Compared with the model group, the expression of CyclinB1 in the 14th and 28th days was significantly different (P <0.05) after intervention with Yiqi Huoxue prescription. Conclusion: Yiqi Huoxue Decoction significantly inhibits the proliferation of vascular intima VSMCs after balloon injury. The mechanism may be related to down-regulating the expression of cyclin B1 and inhibiting the cell cycle progression of VSMCs.
