目的研究关节腔注射Nell-1对于骨关节炎状态下的颞下颌关节软骨是否有保护作用。方法对24只新西兰白兔行双侧颞下颌关节盘部分切除术建立骨关节炎模型。4周后,于所有白兔右侧关节注射Nell-1作为实验组,左侧关节注射同剂量生理盐水作为对照组。分别于注射后12周和24周各随机处死12只白兔,通过组织学观察和逆转录酶-聚合酶链式反应(RT-PCR)检测评价Nell-1对关节软骨的保护作用。结果所有白兔的颞下颌关节均出现了骨关节炎样的组织学病变,但在实验选择的时间节点上,实验组关节软骨的病变程度较对照组轻微,注射后12周实验组Ⅱ型胶原与蛋白聚糖的mRNA表达明显高于对照组,但注射后24周实验组与对照组在蛋白聚糖及白介素1的mRNA表达上无明显差异。结论本研究表明关节腔注射Nell-1或可成为治疗颞下颌关节骨关节炎中软骨退变的一种新选择。
Objective To study whether intra-articular injection of Nell-1 has protective effects on temporomandibular joint cartilage in osteoarthritis. Methods Twenty-four New Zealand white rabbits underwent bilateral partial temporomandibular joint disc resection to establish osteoarthritis model. After 4 weeks, Nell-1 was injected into the right joint of all rabbits as the experimental group, and the left joint was injected with the same dose of saline as the control group. Twelve rabbits were randomly sacrificed at 12 weeks and 24 weeks after injection, respectively. The protective effect of Nell-1 on articular cartilage was evaluated by histological observation and reverse transcriptase-polymerase chain reaction (RT-PCR). Results All the rabbits had osteoarthritis-like histological changes in the temporomandibular joint. However, the lesion degree of articular cartilage in the experimental group was slightly lower than that of the control group at the selected time points. After 12 weeks of injection, the type Ⅱ collagen And proteoglycan mRNA expression was significantly higher than the control group, but 24 weeks after injection experimental group and control group in the proteoglycan and interleukin 1 mRNA expression no significant difference. Conclusions This study shows that intra-articular injection of Nell-1 may be a new option for treating cartilage degeneration in temporomandibular joint osteoarthritis.
