目的 研究腺病毒介导的人 GM- CSF基因转染瘤苗体内抗肿瘤免疫作用极其机理。方法 应用 GM- CSF基因转染、辐照的瘤苗对小鼠肝癌模型进行免疫基因治疗 ,观察该疗法对荷瘤小鼠脾细胞 NK、L AK、CTL 活性的影响、脾淋巴细胞转化反应的影响以及荷瘤小鼠的生存期。结果 经 GM- CSF基因转染瘤苗治疗后 ,脾细胞 CTL活性显著升高 ,而 NK、L AK细胞活性未见明显增强、脾淋巴细胞转化反应显著增强、生存期显著延长。结论 GM- CSF基因转染瘤苗能诱导出有效的抗肿瘤免疫反应 ,揭示了应用该疗法进行肿瘤基因治疗的可行性。
Objective To study the mechanism of anti-tumor immunity of adenovirus-mediated human GM-CSF gene transfection in vivo. Methods GM-CSF gene-transfected and irradiated tumor vaccine was used to immunostaining mouse model of hepatocellular carcinoma. The effects of this therapy on the activity of NK, L AK and CTL in spleen cells of mice with tumor were observed. The spleen lymphocyte transformation reaction Effects and survival of tumor-bearing mice. Results After treatment with GM-CSF gene, the CTL activity of spleen cells was significantly increased, while the activity of NK and L AK cells was not significantly increased. The splenic lymphocyte transformation reaction was significantly enhanced and the survival time was significantly prolonged. Conclusion GM-CSF gene transfection vaccine can induce an effective anti-tumor immune response, revealing the feasibility of using the therapy for gene therapy of cancer.
