目的探讨多巴胺、硝酸甘油诱导的血管舒缩异常大鼠血神经肽和中脑导水管周围灰质c-fos的表达。方法大鼠随机分为溶媒组、多巴胺组、模型组,造模后4h取血放射免疫测降钙基因相关肽(CGRP)、内皮素(ET)、P物质(SP)、β-内啡肽(β-EP),取脑、切片免疫组织化学观察中脑导水管周围灰质(PAG)c-fos蛋白表达。结果偏头痛模型组ET、SP含量较溶媒组明显升高,β-EP含量较溶媒组明显降低,模型组和多巴胺组c-fos阳性细胞面积明显大于溶媒组。结论 多巴胺、硝酸甘油诱发的血管舒缩模型在一定程度上模拟了偏头痛的发病机制。
Objective To investigate the expression of neuropeptide and c-fos in periaqueductal gray of rats with abnormal vasomotor induced by dopamine and nitroglycerin. Methods The rats were randomly divided into vehicle group, dopamine group and model group. Blood samples were collected at 4 hours after the model was given for radioimmunoassay of calcitonin gene related peptide (CGRP), endothelin (ET), substance P (SP), β-endorphin (β-EP). The expression of c-fos protein in midbrain periaqueductal gray (PAG) was observed by immunohistochemistry. Results The content of ET and SP in migraine model group was significantly higher than that in vehicle group and the content of β-EP was significantly lower than that in vehicle group. The area of c-fos positive cells in model group and dopamine group was significantly larger than that in vehicle group. Conclusions The model of vasomotor induced by dopamine and nitroglycerin simulates the pathogenesis of migraine to a certain extent.
