目的:在核磁共振谱仪上建立对同一大鼠心脏机械功能与能量代谢同时测定的技术方法。方法:制备离体等容大鼠心脏模型,通过建立计算机心功能监测系统测量和分析心脏的血流动力学参数;利用心脏的磷-31核磁共振(31P-NMR)谱观测心肌组织的能量代谢。结果:在所建NMR等容心脏模型上,心脏的机械和代谢功能可以实时、动态观测,且在实验结束后即可得到整个实验过程中心脏的血流动力学数据和心肌能量代谢的数据;离体大鼠心脏在NMR实验条件下具有较好的稳定性。结论:所建立的对等容大鼠心脏同时进行NMR和血流动力学观测的实验模型为国内基础心血管医学的研究提供了先进的技术手段。
OBJECTIVE: To establish a method for the simultaneous determination of mechanical and energy metabolism in the same rat heart using nuclear magnetic resonance spectroscopy. Methods: The rat heart model was prepared in vitro. The cardiac hemodynamic parameters were measured and analyzed by establishing a computerized cardiac function monitoring system. The myocardial tissue energy metabolism was measured by using the 31P-NMR spectroscopy . Results: The built-in NMR isokinetic heart model can be used to observe the mechanical and metabolic functions of the heart in real time and dynamically. At the end of the experiment, the hemodynamic parameters of the heart and myocardial energy metabolism data were obtained. Isolated rat heart has good stability under NMR experimental conditions. CONCLUSION: The established experimental model for the simultaneous NMR and hemodynamic observation of isokinetic rat heart provides advanced technical means for the study of basic cardiovascular medicine in China.
