目的 探讨体外药敏试验 (MTT法 )对急性白血病的治疗指导作用及P -gp和CD34抗原表达与白血病治疗疗效的关系。方法 用MTT法对 43例急性白血病患者进行白血病细胞体外药敏试验 ,同时用流式细胞仪检测白血病细胞的P -gp和CD34抗原的表达率。结果 体外药敏与临床疗效的阳性符合率为 77.8% ,阴性符合率为 5 6 .2 %。P -gp+ 、CD+3 4者的临床疗效及体外药敏所占的百分率分别显著低于P -gp- 、CD-3 4者。结论 同时进行MTT法体外药敏试验及P -gp和CD34抗原表达的检测 ,能提高临床耐药判断的准确性 ,有利于合理选用化疗药物以提高疗效
Objective To investigate the therapeutic effect of MTT on acute leukemia and the relationship between the expression of P-gp and CD34 antigen and the therapeutic effect of leukemia. Methods Thirty-four patients with acute leukemia were treated with MTT assay for leukemic cells in vitro. The expression of P-gp and CD34 in leukemia cells was detected by flow cytometry. Results In vitro susceptibility and clinical efficacy of the positive coincidence rate was 77.8%, the negative coincidence rate of 56.2%. The clinical efficacy of P-gp +, CD + 34 and the percentage of drug susceptibility in vitro were significantly lower than those of P-gp- and CD-34, respectively. Conclusions Simultaneous MTT assay in vitro and detection of P-gp and CD34 antigen expression can improve the accuracy of clinical resistance judgment, which is conducive to the rational selection of chemotherapy drugs to improve the curative effect
