目的观察氧化铁磁流体热疗(MFH)对人良性脑膜瘤细胞增殖和凋亡的影响。方法15例人脑膜瘤细胞短期原代培养,进行氧化铁磁流体单次加热或3次加热,加热温度42~44℃,持续30min。流式细胞仪检测细胞凋亡,四氮唑蓝(MTT)比色法检测细胞增殖的变化。结果MFH(44℃,30min)单次加热和3次加热组中,人脑膜瘤细胞增殖抑制率分别为(12.62±0.78)%和(12.78±1.56)%,与对照组相比差异显著(P<0.05)。MFH引起部分脑膜瘤细胞裂解坏死,散在细胞凋亡,但各组细胞凋亡率无明显差异。结论在人良性脑膜瘤体外实验中,氧化铁MFH(44℃,30min)可以明显抑制细胞增殖,但不能有效诱发细胞凋亡。
Objective To observe the effects of ferrofluidic hydro-therapy (MFH) on the proliferation and apoptosis of human meningioma cells. Methods Fifteen human meningioma cells were cultured for short term. The ferrofluid was heated for one time or three times. The temperature was 42 ~ 44 ℃ for 30 min. Cell apoptosis was detected by flow cytometry. Cell proliferation was detected by MTT assay. Results The inhibitory rates of human meningioma cell proliferation were (12.62 ± 0.78)% and (12.78 ± 1.56)%, respectively, in MFH (44 ℃, 30min) single heating and three times heating, which were significantly different from those in control group <0.05). MFH caused part of meningioma cell lysis and necrosis, scattered in apoptosis, but no significant difference in the rate of apoptosis in each group. Conclusion MFH (44 ℃, 30min) can inhibit cell proliferation obviously but can not effectively induce cell apoptosis in human meningioma.
