目的研究含白芍中成药逍遥丸对血虚肝郁证大鼠外周血象和下丘脑-垂体-肾上腺轴的影响。方法以辐照结合慢性束缚应激的方法制备大鼠血虚肝郁模型。健康雄性SD大鼠随机分为对照组、模型组、白芍组、逍遥丸组,对照组常规饲养,其余各组单笼饲养。造模第1天开始ig给药,共给药21 d。检测大鼠体质量、外周血象、糖水消耗量,并于给药结束后测定血浆中促肾上腺皮质激素(ACTH)、皮质酮(CORT)、下丘脑促肾上腺皮质激素释放素(CRH)的量。结果与对照组相比,模型组大鼠体质量、外周血象白细胞(WBC)量明显降低(P<0.001),ACTH、CORT、CRH量均明显升高(P<0.01、0.001),糖水消耗实验表明模型组大鼠快感缺失(P<0.001),逍遥丸对上述各项指标具有相应的调节作用(P<0.01、0.001)。结论逍遥丸对外周血象及HPA轴的调整作用可能是其发挥疏肝解郁、养血健脾功效的机制之一。
Objective To study the effect of Xiaoyao Pills containing Chinese herb peony root on peripheral blood and hypothalamus-pituitary-adrenal axis in rats with deficiency of blood and liver syndrome. Methods The model of blood deficiency and liver depression in rats was prepared by irradiation combined with chronic restraint stress. Healthy male SD rats were randomly divided into control group, model group, Baishao group, Xiaoyao Pill group and control group. The other groups were housed in single cage. The first day of model ig administration, a total of 21 d. The body weight, peripheral blood and sugar consumption of rats were measured. The amount of adrenocorticotropic hormone (ACTH), corticosterone (CORT) and corticotropin releasing hormone (CRH) in the hypothalamus were measured at the end of the administration. Results Compared with the control group, the body weight, the amount of peripheral white blood cells (WBC) and the amount of ACTH, CORT and CRH in the model group were significantly decreased (P <0.01 and 0.001) The rats in model group showed loss of pleasure (P <0.001), and Xiaoyao Pill had corresponding regulation on the above indexes (P <0.01, 0.001). Conclusion The adjustment effect of Xiaoyao Pills on peripheral blood and HPA axis may be one of the mechanisms by which Xiaoyao Pill exerts the effect of relieving stagnation of liver,
