目的观察加味鳖甲煎丸对四氯化碳(CCl4)所致肝纤维化大鼠的治疗作用。方法选用Wistar大鼠由皮下注射40%CCl4溶液首次5 mL/kg,以后每周2次,每次3 mL/kg,连续注射6周,在病理检查证明造模大鼠肝组织出现肝纤维化后,分为模型组、秋水仙碱对照组、肝脾康对照组和加味鳖甲煎丸大、中、小剂量组。连续给药8周后,观察加味鳖甲煎丸对CCl4所致肝纤维化大鼠血清谷草转氨酶(AST)、谷丙转氨酶(ALT)、血清总蛋白(TP)、血清白蛋白(ALB)、血胆红素(TBIL)、白蛋白与球蛋白比值(A/G)、胶原Ⅳ(CⅣ)、层黏蛋白(LN)、透明质酸(HA)含量。结果同模型组相比,加味鳖甲煎丸大、中剂量组可使CCl4造模大鼠血清AST、ALT、TBIL、CⅣ、LN、HA明显降低。结论加味鳖甲煎丸对CCl4所致肝纤维化大鼠有较好治疗作用。
Objective To observe the therapeutic effect of Jiawei Biejiajian Pill on carbon tetrachloride (CCl4)-induced liver fibrosis in rats. Methods Wistar rats were injected subcutaneously with 5 mL/kg of 40% CCl4 solution for the first time, followed by 3 mL/kg twice a week for 6 weeks. Liver fibrosis in the liver of model rats was confirmed by pathological examination. After that, they were divided into model group, colchicine control group, liver-spleen-contrast control group, and Jiawei Biejia Jianwan large-, medium-, and small-dose groups. After 8 weeks of continuous administration, the serum of Aspartate aminotransferase (AST), alanine aminotransferase (ALT), serum total protein (TP), and serum albumin (ALB) in rats with liver fibrosis induced by CCl4 were observed. Blood bilirubin (TBIL), albumin and globulin ratio (A/G), collagen IV (CIV), laminin (LN), hyaluronic acid (HA) content. Results Compared with the model group, the serum AST, ALT, TBIL, CIV, LN, and HA in the CCl4 model rats were significantly decreased in the Jiawei Biejia Jianwan Dazhong and Zhongdu groups. Conclusion Jiawei Biejiajianwan has a good therapeutic effect on CCl4-induced liver fibrosis in rats.
