目的 :研究郑氏植物蛋白 (Zheng’splant protein ,ZPP)对淋巴细胞的刺激作用及其在乳腺癌早期诊断与治疗中的应用价值。方法 :采用3 H TdR掺入法测定人外周血淋巴细胞(PBL) )的增殖反应。用流式细胞仪测定ZPP对T淋巴细胞CD3+ 、CD4 + 和CD8+ 抗原表达的调节作用。结果 :ZPP对乳腺癌及其前期病变患者PBL有较强的刺激作用 (SI分别为3.0 0± 1.4 5和 2 .5 3± 0 .80 ) ,明显高于对正常对照组和乳管上皮轻度增生患者PBL的刺激指数 (SI分别为 1.0 6± 0 .17和1.18± 0 .19) (P <0 .0 1)。而PHA对各组PBL的增殖反应均有不同程度的刺激作用 ,但各组间无明显差异 (P >0 .0 5 ) .经ZPP刺激后的PBL中CD3+ 、CD4 + 和CD8+ 细胞均发生增殖反应 ,其中CD4 + 和CD8+ 者更为明显。结论 :ZPP可选择性刺激乳腺癌及其前期病变患者的PBL中T细胞发生增殖反应 ,此作用有可能用于乳腺癌的早期诊断与治疗中
Objective: To study the effects of Zheng’s plant protein (ZPP) on lymphocyte stimulation and its value in the early diagnosis and treatment of breast cancer. Methods: Proliferation response of human peripheral blood lymphocytes (PBLs) was determined by 3 H TdR incorporation assay. The regulation of ZPP on the expression of CD3 +, CD4 + and CD8 + T lymphocytes by flow cytometry. Results: ZPP had strong stimulating effect on PBL of breast cancer and its pre-lesion (SI = 3.0 0 ± 1.4 5 and 2.53 ± 0.80 respectively), which was significantly higher than that of normal control group and ductal epithelium Proliferation of patients with PBL stimulation index (SI respectively 1.06 ± 0.17 and 1.18 ± 0.19) (P <0.01). However, the proliferative responses of PBLs in different groups were all stimulated by PHA, but there was no significant difference among groups (P> 0.05) .P3 +, CD4 + and CD8 + cells were proliferated in PBL stimulated by ZPP Reaction, which CD4 + and CD8 + were more obvious. Conclusion: ZPP can selectively stimulate the proliferation of T cells in PBL of breast cancer and its pre-lesion, which may be used in the early diagnosis and treatment of breast cancer
