目的研究厄洛替尼对单核细胞分化发育过程中核转录因子(NF)κB表达和凋亡的影响。方法分别在单核细胞系的培养液中加入EGF、厄洛替尼、EGF+厄洛替尼处理,Western印迹检测NF-κB的表达情况,流式细胞学检测单核细胞的凋亡率。结果厄洛替尼对人单核细胞的凋亡具有抑制作用,其抑制作用具有明显的剂量和时间依赖性(P<0.05);各组单核细胞的凋亡率差异有统计学意义(P<0.05);厄洛替尼可以显著地抑制单核细胞分泌NF-κB。结论厄洛替尼可以有效抑制单核细胞凋亡和迁移,进而调控单核细胞的分化。 Objective To investigate the effect of erlotinib on the expression of nuclear factor κB (NF-κB) and apoptosis during monocyte differentiation and development. Methods EGF, erlotinib and EGF + erlotinib were respectively added into the culture of mononuclear cell line. The expression of NF-κB was detected by Western blotting. The apoptosis rate of monocytes was detected by flow cytometry. Results Erlotinib inhibited the apoptosis of human mononuclear cells in a dose-and time-dependent manner (P <0.05). The apoptotic rates of mononuclear cells in each group were significantly different (P <0.05). Erlotinib can significantly inhibit monocytes secreting NF-κB. Conclusion Erlotinib can effectively inhibit monocyte apoptosis and migration, and then regulate monocyte differentiation.
