目的探讨铀矿尘致大鼠肺组织纤维化对血浆中Ⅲ型前胶原氨端肽(PⅢNP)、层粘连蛋白(LN)和转化生长因子-β1(TGF-β1)的影响。方法 60只大鼠随机分为铀矿尘组和对照组,铀矿尘组每只大鼠气管内一次性滴注20 mg/mL粉尘悬液1 mL,对照组给予等量生理盐水,分别于处理后第7、14、21、30及60 d采取肺组织及外周血,进行肺组织形态学观察及血浆中PⅢNP、LN和TGF-β1含量测定。结果在染尘后第21、30、60 d时,PⅢNP含量为(17.90±1.55)、(19.06±1.62)、(20.09±1.65)ng/mL,LN含量为(81.81±1.32)、(83.64±1.52)、(84.73±2.20)ng/mL,TGF-β1含量为(34.03±3.21)、(46.65±3.09)、(40.95±3.18)pg/mL,与对照组比较,差异有统计学意义(P<0.01),血浆中PⅢNP和LN含量增高与肺组织纤维化程度具有较好一致性。结论铀矿尘诱导大鼠肺组织纤维化过程中可引起血浆中PⅢNP、LN和TGF-β1含量明显增高。
Objective To investigate the effects of lung tissue fibrosis induced by uranium mine dust on plasma type Ⅲ procollagen (PⅢNP), laminin (LN) and transforming growth factor-β1 (TGF-β1) in plasma. Methods Sixty rats were randomly divided into uranium dust group and control group. In the uranium mine dust group, 1 mL of 20 mg / mL dust suspension was injected into the trachea of each rat. The control group was given the same amount of normal saline, Pulmonary tissues and peripheral blood were collected on the 7th, 14th, 21st, 30th and 60th day after treatment. Morphology of lung tissue and contents of PⅢNP, LN and TGF-β1 in plasma were determined. Results The levels of PⅢNP were (17.90 ± 1.55), (19.06 ± 1.62) and (20.09 ± 1.65) ng / mL on the 21th, 30th and 60th day after dying. The LN contents were 81.81 ± 1.32 and 83.64 ± (P <0.05). Compared with the control group, the difference was statistically significant (P <0.05), while the level of TGF-β1 was (34.03 ± 3.21), (46.65 ± 3.09) <0.01). The plasma levels of PⅢNP and LN were in good agreement with the degree of lung fibrosis. Conclusion Uranium dust-induced pulmonary fibrosis in rats can cause plasma PⅢNP, LN and TGF-β1 levels were significantly increased.
